fa بهینه‌سازی درمانی آپی‌ژنین در سرطان کولورکتال مرتبط با کولیت؛ کاهش استرس اکسیداتیو و التهاب در یک دوز مؤثر غیرسمی در مدل موشی AOM/DSS: یک مطالعه تجربی Therapeutic Optimization of Apigenin in Colitis-Associated Colorectal Cancer: Attenuation of Oxidative Stress and Inflammation at an Effective Non-Toxic Dose in an AOM/DSS Mouse Model انكولوژي Oncology پژوهشي Research زمینه و هدف: استرس اکسیداتیو در سرطان کولون نقش دارد. آپیژنین، فلاونوئیدی با اثرات آنتی‌اکسیدانی قوی است که در درمان سرطان مؤثر است. این مطالعه با هدف تعیین اثر درمانی آپیژنین بر سرطان کولون بهروش مکانیسم‌های آنتی‌اکسیدانی انجام شد. مواد و روش‌ها: مطالعه حاضر از نوع تجربی است و در این مطالعه تعداد ۶۰ سر موش نر نژاد C57BL/6 به شش گروه کنترل سالم، کنترل بیمار و چهار گروه درمانی که سرطان‌زایی کولون در آن‌ها با تزریق داخل صفاقی ۱۰ میلی‌گرم/کیلوگرم آزوکسی‌متان (Azoxymethane; AOM) و دو دوره 5/2 درصد سدیم دکسترانسولفات (Sodium Dextran Sulfate; DSS) القاء شد. سپس دوزهای ۵، ۱۰، ۲۰ و ۴۰ میلی‌گرم/کیلوگرم آپی‌ژنین به‌صورت خوراکی و با تناوب سهبار در هفته به‌مدت شش هفته تجویز شد. نشانگرهای استرس اکسیداتیو شامل گونه‌های فعال اکسیژن (Reactive Oxygen Species; ROS)، مالون‌دی‌آلدئید (Malondialdehyde; MDA)، سوپراکسید دیس موتاز (Superoxide Dismutase; SOD)، ظرفیت آنتی‌اکسیدانی کل (Total Antioxidant Capacity; TAC)، گلوتاتیون پراکسیداز (Glutathione Peroxidase; GPx) و کاتالاز بررسی شد. داده‌ها با استفاده از آنالیز واریانس یک‌طرفه و آزمون تعقیبی Tukey تجزیهوتحلیل شدند. یافته‌ها: در دوز ۲۰ میلی‌گرم/کیلوگرم نسبت به دوزهای 5، 10، و 40 میلیگرم/کیلوگرم آپیژنین کاهش معنی‌دار ROS و MDA و افزایش فعالیت SOD، GPx، کاتالاز و TAC مشاهده شد (۰۵/۰>P). نتیجه‌گیری: آپی‌ژنین در مدل حیوانی سرطان کولورکتال، استرس اکسیداتیو را کاهش و دفاع آنتی‌اکسیدانی را تقویت کرد و دوز ۲۰ میلی‌گرم/کیلوگرم کمترین سمیت کبدی را نشان داد. تعمیم این نتایج به انسان نیازمند مطالعات بالینی تکمیلی است. گنجاندن منابع غذایی حاوی آپی‌ژنین در رژیم غذایی می‌تواند به‌عنوان راهکار پیشگیرانه کمکی مطرح شود. واژه‌های کلیدی: آپی‌ژنین، سرطان کولون، استرس اکسیداتیو   <a name="_Hlk114425361">Background and Objectives:</a> Oxidative stress plays a role in the pathogenesis of colon cancer. Apigenin, a flavonoid with potent antioxidant properties, is effective in cancer treatment. The present study aimed to determine the therapeutic effect of apigenin on colon cancer through antioxidant mechanisms. Materials and Methods: This is an experimental research, in which 60 male C57BL/6 mice were divided into six groups, including a healthy control group, a diseased control group, and four treatment groups in which colon carcinogenesis was induced by intraperitoneal injection of 10 mg/kg azoxymethane (AOM) and two cycles of 2.5% sodium dextran sulfate (DSS). Subsequently, doses of 5, 10, 20, and 40 mg/kg of apigenin were administered orally three times per week for six weeks. Oxidative stress markers including reactive oxygen species (ROS), malondialdehyde (MDA), superoxide dismutase (SOD), total antioxidant capacity (TAC), glutathione peroxidase (GPx), and catalase were evaluated. Data were analyzed using one-way analysis of variance (ANOVA) and Tukey's post hoc test. Results: At a dose of 20 mg/kg of apigenin, a significant decrease in ROS and MDA and a significant increase in the activity of SOD, GPx, catalase, and TAC were observed (P<0.05). Conclusion: Apigenin, in an animal model of colorectal cancer, reduced oxidative stress and enhanced antioxidant defense, and the dose of 20 mg/kg exhibited the lowest hepatic toxicity. Generalization of these results to humans requires further clinical studies. Incorporating dietary sources containing apigenin into  diet may be proposed as an adjunctive preventive strategy.
