fa اثر ضد قارچی عصاره‌های آبی و متانولی برگ گیاه حرا (Avicennia marina) بر آلترناریا آلترناتا و پنی‌سیلیوم سیترینوم Antifungal Effect of Aqueous and Methanolic Avicennia Marina Leaves Extracts on Alternaria Alternata and Penicillium Citrinum ميكروبيولوژي Microbiology پژوهشي Research  gte vml 1]><v:roundrect id="_x0000_s1026" style='position:absoluteleft:0text-align:leftmargin-left:2.45pt margin-top:1.4ptwidth:449.5ptheight:397.15ptz-index:-1' arcsize="1475f"> <w:wrap anchorx="page"/> </v:roundrect><img alt="AWT IMAGE" width="602" height="531" src="file:///C:/DOCUME~1/ADMINI~1/LOCALS~1/Temp/msohtmlclip1/01/clip_image001.gif" v:shapes="_x0000_s1026" >چکیده<o:p /> زمینه و هدف: خانواده Avicenniaceae یکی از اعضای گیاهان مانگرو حقیقی است که دارای یک جنس، یازده گونه و چند زیرگونه می‌باشد. با توجه به وجود ترکیبات بیولوژیکی فعال و استفاده‌هایی که از این گیاه در طب سنتی به عمل می‌آید، به نظر می‌رسد این گیاه دارای اثرات ضد قارچی قابل ملاحظه‌ای باشد. هدف از این مطالعه، بررسی اثر ضد قارچی عصاره‌های آبی و متانولی برگ گیاه حرا علیه قارچ آلترناریا آلترناتا و پنی‌سیلیوم سیترینوم می‌باشد.<o:p /> مواد و روش‌ها: در این پژوهش آزمایشگاهی، اثر ضد قارچی عصاره به دو روش تمام ظرف و روش انتشار در آگار بر میکروارگانیسم‌های <a href="http://www.irost.org/persian/ptcc/DBank-details.asp?id=371&code=1">Alternaria alternata PTCC 5224</a> و <a href="http://www.irost.org/persian/ptcc/DBank-details.asp?id=545&code=1">Penicillium citrinum PTCC 5304 </a>مورد بررسی قرار گرفت. حداقل غلظت مهارکنندگی (MIC)و حداقل غلظت کشندگی (MFC) با استفاده از روش رقت لوله‌ای تعیین شد. تجزیه و تحلیل نتایج با استفاده از آزمون آنالیز واریانس (ANOVA) انجام شد. <o:p /> یافته‌ها: در روش انتشار در آگار همه غلظت‌های عصاره متانولی اثر بازدارندگی داشت. در روش تمام ظرف عصاره متانولی در غلظت 2 میلی‌گرم بر میلی‌لیتر اثر بازدارندگی داشت، اما عصاره آبی در غلظت 2 میلی‌گرم بر میلی‌لیتر فاقد اثر ضد قارچی مشخصی بود. MICعصاره متانولی برگ حرا برای پنی‌سیلیوم 8 میلی‌گرم بر میلی‌لیتر و برای آلترناریا 16 میلی‌گرم بر میلی‌لیتر بود، در حالی که MIC عصاره آبی برای پنی‌سیلیوم 32 میلی‌گرم بر میلی‌لیتر و برای آلترناریا 64 میلی‌گرم بر میلی‌لیتر بود. MFC عصاره متانولی برگ حرا برای پنی‌سیلیوم 16 میلی‌گرم بر میلی‌لیتر و برای آلترناریا 32 میلی‌گرم بر میلی‌لیتر بود در حالی که MFC عصاره آبی برای پنی سیلیوم 64 میلی‌گرم بر میلی‌لیتر و برای آلترناریا 256 میلی‌گرم بر میلی‌لیتر بود. <o:p /> نتیجه‌گیری: عصاره متانولی برگ گیاه حرا در مقایسه با عصاره آبی در شرایط آزمایشگاهی اثر بازدارندگی بیشتری بر روی سویه‌های مورد مطالعه داشت. <o:p /> واژه‌های کلیدی: گیاه حرا، عصاره آبی و متانولی، اثرضد قارچی<o:p /> Background and Objective: Avicenniaceae family is a member of true mangrove plants which has one genus, 11 species and several sub species. According to biologically active compounds and traditional uses of the leaves in the traditional medician it, seems that this plant has significant anti-fungal effects. The aim of this study was evaluation of antifungal effect of aqueous and methanolic extracts of Avicennia marina leaves against Alternaria alternata and Penicillium citrinum.<o:p /> Materials and Methods: In this labratory study antifungal effect were of extracts evaluated by two methods, “Collins method” and “disk agar diffusion method” on Alternaria alternata PTCC 5224 and Penicillium citrinum PTCC 5304 microorganisms. The minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) for both species were determined using “dilution method”. Statistical analysis was carried out using analysis of variance (ANOVA) test.<o:p /> Results: All methanolic extract concentrations had inhibitory effect in the disk agar diffusion method. In “Collins method” 2 mg/ml concentration of methanolic extract could prevent the growth of both strains on medium. The aqueous extract had no antifungal significant effect in 2 mg/ml concentration. The methanolic extract MIC of Avicennia marina leaves for Penicillium citrinum and Alternaria alternata were respectively 8 mg/ml and 16 mg/ml. But the aqueous extracts MIC of Avicennia marina leaves for Penicillium citrinum was 32 mg/ml and 64 mg/ml for Alternaria alternata was. The methanolic extract MFC of Avicennia marina leaves for Penicillium citrinum and Alternaria alternata were respectively 16 mg/ml and 32 mg/ml but the aqueous extracts MFC of Avicennia marina leaves for Penicillium citrinum was 64 mg/ml and 256 mg/ml for Alternaria alternata.<o:p /> Conclusions: The methanolic extract of Avicennia marina leaves had more inhibitory effect than the aqueous extracts on both fungal species. <o:p /> Keywords: Avicennia marina, Aqueous and methanolic extracts, Antifungal effect <o:p />   Funding: This research was funded by Department of Food Science and Technology, Faculty of Agriculture, Ferdowsi University of Mashhad.<o:p /> Conflict of interest: None declared.<o:p /> Ethical approval: The Ethics Committee of Ferdowsi University of Mashhad approved the study.<o:p />   How to cite this article: Alizadeh Behbahani B, Tabatabaei Yazdi F, Shahidi F, Mohebbi M.
