fa اثر ضد اسپاسمی فراکسیون متانولی مشتق از اسانس بادیان رومی (Trachyspermum Ammi) بر انقباضات ایلئوم موش صحرایی The Antispasmodic Effect of Metanolic Fraction Derived from Trachyspermum Ammi Essense on Rat's Ileum Contraction فيزيولوژي Physiology پژوهشي Research  gte vml 1]><v:roundrect id="_x0000_s1026" style='position:absolute left:0text-align:leftmargin-left:2.45ptmargin-top:1.4ptwidth:449.5pt height:411.15ptz-index:-1' arcsize="1475f"> <w:wrap anchorx="page"/> </v:roundrect><img alt="AWT IMAGE" width="602" height="550" src="file:///C:/DOCUME~1/ADMINI~1/LOCALS~1/Temp/msohtmlclip1/01/clip_image001.gif" v:shapes="_x0000_s1026" >چکیده<o:p /> زمینه و هدف: استفاده از گیاهان دارویی، از دیرباز در طب سنتی برای معالجه بیماری‌های مختلف از جمله اختلالات گوارشی رایج بوده است. یکی از این گیاهان که در طب سنتی ایران از آن یاد شده بادیان رومی است که تحقیقات زیادی در مورد اثرات فارماکولوژیک آن انجام شده است. مطالعه حاضر، نقش فراکسیون متانولی مشتق از این اسانس را بر فعالیت مکانیکی عضلات صاف ایلئوم جدا شده از موش صحرایی مورد بررسی قرار داده است.<o:p /> مواد و روش‌ها: این مطالعه آزمایشگاهی در دانشکده پزشکی دانشگاه علوم پزشکی یزد بخش فیزیولوژی در ابتدای سال 1392 انجام گرفت. جهت بررسی آثار اسپاسمولیتیک فراکسیون، محلول‌های مختلف را پس از اعمال استیل‌کولین (Ach)4-10 مولار، به حمام بافت اضافه کردیم و همچنین، جهت بررسی آثار آنتی‌اسپاسمودیک فراکسیون، محلول‌های مختلف را قبل از اعمال استیل‌کولین (Ach) 9-10تا 2-10 مولار اضافه کردیم و سپس با استفاده از دستگاه فیزیوگراف و حمام بافت، انقباضات ایزوتونیک ایلئوم مورد بررسی قرار گرفت. نتایج حاصل با اثرات تیمول به‌عنوان کنترل مثبت مقایسه گردید.<o:p /> یافته‌ها: نتایج بدست آمده از اثر غلظت‌های مختلف این فراکسیون 002/0 ، 005/0، 01/0% برانقباض ناشی از استیل‌کولین 4-10 مولار در ایلئوم موش نشان داد غلظت 002/0% اسانس و غلظت بالاتر آن منجر به کاهش انقباض ناشی از استیل‌کولین می شود. به طوری که غلظت 002/0% آن 8/59% (05/0p<) و غلظت 01/0% آن شدت انقباضات ناشی از استیل‌کولین را 5/77% (01/0p<) کاهش داد. از طرفی غلظت‌های 5-10 و 4-10 مولار تیمول در مقایسه با فراکسیون متانولی با غلظت 002/0و 005/0 و 01/0% کاهش معنی‌داری بر انقباضات ناشی از استیل‌کولین نشان نداد. <o:p /> نتیجه‌گیری: فراکسیون متانولی مشتق از اسانس بادیان رومی دارای آثار اسپاسمولیتیک و آنتی‌اسپاسمودیک می‌باشد.<o:p /> واژه‌های کلیدی: فراکسیون متانولی، بادیان رومی، ضد اسپاسمی، استیل‌کولین، ایلئوم<o:p /> Background and Objective: Since past time, herbal plants have been used for the treatment of different abnormalities such as gastrointestinal disorders. One of these herbs which used in Iranian traditional medicine is Trachyspermum ammi. There are many biological studies conducted for the determination of its pharmacological properties. The present study was designed to find out the effects of methanolic fraction derived from Trachyspermum ammi essense on mechanical activity of the isolated rat’s ileum. <o:p /> Materials and Methods: In this laboratory study, for evaluation of spasmolythic property of fraction, different doses of the solution were added to organ bath after acetylcholine with concentration of 10-4 molar (M) and for assesment of antispasmodic effect of fraction, different doses of the solution were added to the organ bath before acetylcholine with concentration of 10-9 upto 10-2 M, then isotonic contraction of ileum were recorded through an isolated tissue chamber in an organ bath by using isotonic transducer and oscillographic device. The results were compared with thymol as a positive control. <o:p /> Results: Our findings showed that different concentration of metanolic fraction of Trachyspermum ammi 0.002, 0.005 & 0.01% on acetylcholine 10-4 M induced contraction in rat’s ileum resulted in reduced contraction by the essense from concentration of 0.002% and higher. So that the concentration of 0.002% was reduced to 59.8% (P< 0.05) and concentration of 0.01% was reduced to 77.5% (P< 0.01). in addition thymol in concentration of 10-5 M & 10-4 M as compared with metanolic fraction of 0.002, 0.005 & 0.01% had no significant effect on acetylcholine induced contraction.<o:p /> Conclusion: Methanolic fraction derived from Trachyspermum ammi essense has spasmolythic and antispasmodic property.<o:p /> Key word: Metanolic fraction, Trachyspermum ammi.
