fa اثرات آپاپتوزی آلوئه‌امودین (Aloe-emodin) بررده سلولی MCF-7 Apoptosis Effects of Aloe-emodin against MCF-7 Cell Line بیوشیمی Biochemistry پژوهشي Research  gte vml 1]><v:roundrect id="_x0000_s1026" style='position:absolute left:0text-align:leftmargin-left:2.45ptmargin-top:3.65ptwidth:449.5pt height:339.15ptz-index:-1' arcsize="1475f"> <w:wrap anchorx="page"/> </v:roundrect><img alt="AWT IMAGE" width="602" height="454" src="file:///C:/DOCUME~1/ADMINI~1/LOCALS~1/Temp/msohtmlclip1/01/clip_image001.gif" v:shapes="_x0000_s1026" >چکیده<o:p /> زمینه و هدف: در مطالعات متعددی به خواص بیولوژیکی آلوئه‌امودین شامل: خاصیت ضد ویروسی، ضد میکروبی و اثرات حفاظتی آن بر کبد اشاره شده است. اخیراً خواص ضدتوموری آلوئه‌امودین توجه بسیاری از محققین را به خود جلب کرده است. سرطان پستان شایع‌ترین بدخیمی زنان در سراسر دنیاست. به طوری که 30% از کل سرطان‌ها و 15% از مرگ‌های ناشی از سرطان در میان زنان، و کمتر از 1% سرطان‌ها در مردان مربوط به آن است. هدف از مطالعه حاضر، بررسی اثر ضد سرطانی آلوئه‌امودین بر سلول‌های سرطان پستان، MCF-7 بود.<o:p /> مواد و روش‌ها: در این مطالعه آزمایشگاهی، سلول‌های سرطان پستان (MCF-7) در محیط کشت RPMI با 10% سرم جنین گاوی کشت داده شدند. اثر سایتوتوکسیسیتی غلظت‌های 5، 10، 25، 50 و 100 میکرومول آلوئه‌امودین، بعد از 24، 48 و 72 ساعت انکوبه کردن سلول‌های کشت داده شده، به وسیله آزمون MTT ارزیابی گردید. آنالیز آپاپتوز و بیان Fas(CD95) به وسیله دستگاه فلوسایتومتری با کیت انکسین- پروپیدیوم یدید (Annexin-PI) و کیت بیان فعالیت Fas(CD95)، طبق دستورالعمل کیت انجام گرفت.<o:p /> یافته‌ها: با افزایش غلظت آلوئه‌امودین به صورت وابسته به دوز و زمان، توان زیستی سلول‌ها کاهش پیدا کرد. به طوری که بیشترین تأثیر آلوئه‌امودین مربوط به غلظت 100 میکرو‌مول و در زمان 72 ساعت پس از تیمار سلول‌ها بود. آپاپتوز القا شده و میزان بیان Fas نیز وابسته به دوز بودند. غلظت 100 میکرومول از آلوئه‌امودین بیشترین درصد آپاپتوز و بیشترین میزان بیان فعالیت Fas را در سلول‌های MCF-7 نشان داد.<o:p /> نتیجه‌گیری: یافته‌های مطالعه حاضر نشان داد که آلوئه‌امودین (Aloe-emodin) دارای خواص ضد سرطانی است که می‌تواند در درمان سرطان پستان مؤثر باشد. البته نیاز است تحقیقات بیشتر، به خصوص بررسی‌های درون‌تنی انجام گیرد.<o:p /> واژه‌های کلیدی: آلوئه‌امودین (Aloe-emodin)، سرطان پستان، آپاپتوز، MCF-7<o:p /> Background and Objective: Aloe-emodin is a Hydroxyanthraquinone that exists in Aloe Vera leaves and has been known to have anti-tumor effects. Various studies have mentioned the biological effects of Aloe Vera such as antiviral, antimicrobial and some hepato-protective effects. Recently its antitumor effects have attracted the attention of many researchers. Breast cancer is the most common malignancy in women, accounting for 30% of all female cancers and < 1% of all cancer cases in men. Breast cancer is also responsible for 15% of cancer deaths in women. The aim of the present study was to investigate the anti-cancer effects of aloe-emodin on human breast cancer MCF-7 cell line.<o:p /> Materials and Methods: In this laboratory study MCF-7 cells were cultured in RPMI medium with 10% fetal bovine serum (FBS). The cytotoxic effect of different concentrations (5, 10, 25, 50, and 100 µM) of Aloe-emodinon cultured cells were evaluated by MTT assay at different times (24, 48, 72 h). Apoptosis and CD95 (Fas) expression were analyzed by flow-cytometry using an Annexin V-FITC/PI kit and Fas (CD95) kit according to the manufacturer’s protocol.<o:p /> Results: Aloe-emodin decreases the viability of MCF-7 cell line in a time and dose dependent manner, so that the most effective concentration of this substance was 100µM and 72 h after treatment. According to the data of Fas (CD95) expression and Annexin-PI, the highest apoptosis induction rate was seen in 100µM and 72 h.<o:p /> Conclusion: The findings of the present study indicated that Aloe-emodinhas has some antitumor effects and can be used in the treatment of breast cancer.
