fa بررسی اثـر نانـوذرات نقـره بر بیـوفیلم‌های ناشی از استافیلوکوکوس اپیدرمیدیس Study of the Effect of Silver Nanoparticles on Biofilms Formation by Staphylococcus epidermidis ميكروبيولوژي Microbiology پژوهشي Research  gte vml 1]><v:roundrect id="_x0000_s1026" style="position: absolute left: 0px text-align: left margin-left: 5.45pt margin-top: -0.15pt width: 449.5pt height: 388.55pt z-index: -1 " arcsize="1475f"> <w:wrap anchorx="page"> </w:wrap></v:roundrect><img alt="AWT IMAGE" width="602" height="520" src="file:///C:/DOCUME~1/ADMINI~1/LOCALS~1/Temp/msohtmlclip1/01/clip_image001.gif" v:shapes="_x0000_s1026" >چکیده<o:p /> زمینه و هدف: استافیلوکوکوس اپیدرمیدیس پلی‌ساکارید خارج سلولی تولید می‌کند که به عنوان بیوفیلم شناخته می‌شود. بیوفیلم در ایجاد عفونت‌های این باکتری بسیار مؤثر است و می‌تواند روی وسایل پزشکی که در بدن به کار گرفته می‌شوند، ایجاد شود. هدف از اجرای این تحقیق بررسی اثر نانوذرات نقره کلوئیدی بر رشد باکتری استافیلوکوکوس اپیدرمیدیس و بیوفیلم ناشی از آن بود.<o:p /> مواد و روش‌ها: در این مطالعه مقطعی تجربی، آزمایشات بر روی 13 ایزوله بالینی و یک سویه استاندارد استافیلوکوکوس اپیدرمیدیس (ATCC 12228) انجام شد. حداقل غلظت ممانعت از رشد (MIC) و کشندگی (MBC) نانوذرات با روش رقت در آگار تعیین شد. آزمایشات ایجاد بیوفیلم با روش میکروتیتر پلیت و رنگ آمیزی با سافرانین انجام شد. داده‌ها با استفاده از آنالیز واریانس یکطرفه تجزیه تحلیل شدند.<o:p /> یافته‌ها: میانگین MIC و MBC نانوذرات برای ایزوله‌های بیمارستانی به ترتیب 61/19±350 و ppm05/64‌±‌69/707 بود و نانوذره در غلظت‌های پایین اثر باکتریواستاتیک داشت. نانوذره در غلظت ppm 5/0 اثر ضد بیوفیلمی داشت و با افزایش غلظت تا ppm 4 اثر ضد بیوفیلمی آن افزایش یافت. همچنین، نانوذره در غلظت‌های بالاتر از ppm 150 توانایی از بین بردن بیوفیلم تشکیل شده را داشت. <o:p /> نتیجه‌گیری: نتایج تحقیق نشان داد نانوذرات کلوئیدی نقره توانایی مهار تشکیل بیوفیلم استافیلوکوکوس اپیدرمیدیس را دارند و می‌توان پیشنهاد داد که از این ترکیبات در تهیه روکش‌های پروتزها و وسایل مصنوعی که در بدن تعبیه می‌شوند و یا پوشاندن سطوح مراکز درمانی استفاده شود.<o:p /> واژه‌های کلیدی: استافیلوکوکوس اپیدرمیدیس، بیوفیلم، میکروتیتر پلیت<o:p /> <div class="WordSection1"> <h5 align="center" dir="RTL" style="text-align: justify margin-top: 0cm margin-right: 240.95pt margin-left: 0cm margin-bottom: 0.0001pt line-height: 90% "><div class="WordSection1"> <h5 align="center" dir="RTL" style="text-align: justify margin-top: 0cm margin-right: 240.95pt margin-left: 0cm margin-bottom: 0.0001pt line-height: 90% ">Background and Objective: Staphylococcus epidermidis produces extracellular polysaccharide which is known as a biofilm. Biofilm is highly effective in establishing of this bacterium infections and can be formed on medical devices that are used in the body. The purpose of this study was to evaluate the effects of silver colloidal nanoparticles on bacterial growth and biofilm formation of Staphylococcus epidermidis.<o:p /> Materials and Methods: In this empirical and cross-sectional study, experiments were performed on 13 clinical isolated specimens of Staphylococcus epidermidis and a reference strain (ATCC 12228). The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of nanoparticles were determined by the agar dilution method. The experiments of biofilm formation were done by micro titer plate method and stained by Safranin. Data were analyzed using ANOVA.<o:p /> Results: The mean MIC and MBC of nanoparticles for clinical isolated specimens were 350 ± 19.61 ppm and 707.69 ± 64.05 ppm, respectively and nanoparticles had bacteriostatic effect at low concentrations . Nanoparticles showed the anti-biofilm activity at the concentration of 0.5 ppm and this effect enhanced with increasing concentration up to 4 ppm. Nanoparticles at higher concentrations of 150 ppm had the ability to eliminate the pre-formed biofilm.<o:p /> Conclusion: The results showed that colloidal silver nanoparticles have the ability to inhibit the S. epidermidis biofilm formation.
