fa تأثیر روش استخراج عصاره اندام هوایی گیاه دارویی سرخارگل (Echinacea Purpurea L.) بر خاصیت ضد میکروبی آن در تعدادی از باکتری‌های بیماری‌زا Effect of Extraction Method on Antimicrobial Properties of Shoot Extract of Purple Coneflower (Echinacea Purpurea l.) Against Some Pathogenic Bacteria علوم آزمايشگاهي علوم آزمايشگاهي پژوهشي Research چکیده<o:p /> زمینه و هدف: در سال‌های اخیر بر استفاده از مواد طبیعی مانند اسانس‌ها و عصاره‌ها به جای نگهدارنده‌های شیمیایی در صنعت غذا تأکید بیشتری شده است. هدف اصلی تحقیق حاضر تأثیر روش استخراج عصاره اندام هوایی گیاه دارویی سرخارگل بر خاصیت ضد میکروبی آن در تعدادی از باکتری‌های گرم مثبت و گرم منفی می‌باشد.<o:p /> مواد و روش‌ها: این مطالعه آزمایشگاهی در سال 1391 انجام گرفت. ترکیب‌های فنلی عصاره سرخارگل از طریق روش خیساندن و امواج مایکروویو با حلال‌های آب، متانول 80% و استون استخراج و اندازه‌گیری شد. میکروارگانیسم‌های مورد پژوهش باسیلوس سرئوس، لیستریا مونوسیتوژنز، اشرشیاکلی، شیگلا دیسانتری و سودوموناس آئروژینوزا بودند. تجزیه آماری داده‌های آزمایش با استفاده از نرم‌افزار SPSS نسخه 15 و مقایسه میانگین‌ها به روش آزمون چند دامنه‌ای دانکن انجام شد.<o:p /> یافته‌ها: عصاره متانولی در روش استخراج به کمک مایکروویو بیشترین میزان ترکیب‌های فنلی را به خود اختصاص داد، در حالی که عصاره استونی کمترین میزان ترکیب‌های فنلی را در هر دو روش استخراج شامل شد. در مقایسه دو روش استخراج، عصاره‌های مایکروویوی در مورد هر سه حلال مورد آزمون کارایی استخراج بالاتری داشتند. در بررسی فعالیت ضد میکروبی عصاره اندام هوایی سرخارگل، کمترین غلظت بازدارندگی و کمترین غلظت کشندگی در برابر باکتری‌های گرم مثبت باسیلوس سرئوس و لیستریا مونوسیتوژنز حاصل شد. سودوموناس آئروژینوزا مقاوم‌ترین باکتری نسبت به عصاره‌ها بود. <o:p /> نتیجه‌گیری: نتایج نشان داد که در بیشتر موارد عصاره‌های مایکروویوی نسبت به عصاره‌های استخراج شده با روش خیساندن، فعالیت ضد میکروبی بیشتری داشتند. بنابراین عصاره استخراج شده این گیاه به کمک امواج مایکروویو می‌تواند به عنوان یک نگهدارنده مواد غذایی مطرح شود. با وجود این انجام تحقیقات بیشتر در مدل‌های غذایی، اثرات ضد میکروبی آن را آشکارتر خواهد کرد. <o:p /> <div style="text-align: right ">واژه‌های کلیدی: عصاره سرخارگل، خواص ضد میکروبی، روش عصاره‌گیری</div> Background and Objective: In recent years, it is recommended to use natural materials like plant extracts and essences instead of chemical preservatives in food industry. The main objective of this study was to evaluate the antimicrobial activity of shoot extract of purple coneflower against some gram positive and gram negative bacteria. <o:p /> Materials and Methods: This Laboratory study was conducted in 2013. Two extraction methods (maceration and microwave-assisted) with three solvents (water, methanol 80% and acetone) were used to extract the phenolic compounds from purple coneflower shoot The micro-organisms investigated in this study were: Bacillus cereus, Listeria monocytogenes, Escherichia coli, Shigella dysenteriae and Pseudomonas aeruginosa. Experimental data were analyzed using ANOVA by the SPSS version 15 software and mean comparison were done using the Duncan's multiple range test.<o:p /> Results: The highest total phenolic content was related to the methanol extract produced by microwave-assisted extraction (MAE), whereas in both extraction methods, the lowest amount of phenolic was obtained by using acetone extract. Comparing the extraction methods showed that MAE had higher extraction efficiency in all three tested solvents. Regarding to antimicrobial activity of purple coneflower shoot extracts, the minimum inhibitory concentration and minimum bactericidal concentration were observed in gram positive bacteria (Bacillus cereus and Listeria monocytogenes). Pseudomonas aeruginosa was the most resistant bacterium against the extracts. <o:p /> Conclusion: The results showed that in most cases, the extracts obtained by MAE method had more antimicrobial activity in comparison to traditional method. In addition the microwave-assisted extract of this plant can be considered as a food preservative.
