fa بررسی تأثیر ویتامین D3 و عصاره زنجبیل بر علایم بالینی و شدت التهاب در بیماری آنسفالومیلیت خود ایمن تجربی Evaluation of the Effect of Vitamin D3 and Ginger Extract on the Clinical Symptoms and the Severity of Inflammation in EAE فارماكولوژي Pharmacology پژوهشي Research  gte vml 1]><v:roundrect id="_x0000_s1026" style="position: absolute left: 0px text-align: left margin-left: 2.45pt margin-top: 1.4pt width: 449.5pt height: 327.1pt z-index: -1 " arcsize="1475f"> <w:wrap anchorx="page"> </w:wrap></v:roundrect><img alt="AWT IMAGE" width="602" height="438" src="file:///C:/DOCUME~1/ADMINI~1/LOCALS~1/Temp/msohtmlclip1/01/clip_image001.gif" v:shapes="_x0000_s1026" >چکیده<o:p /> زمینه و هدف: بیماری آنسفالومیلیت خودایمن تجربی(EAE) با ارتشاح لکوسیتی سیستم عصبی مرکزی همراه می‌باشد. مطالعات نشانگر اثرات ضد التهابی ویتامین D3 و زنجبیل می‌باشد. هدف این مطالعه بررسی اثر ویتامینD3 و عصاره زنجبیل بر روی علایم بالینی EAE و میزان ارتشاح لکوسیتی است.<o:p /> مواد و روش‌ها: مطالعه حاضر از نوع تجربی می‌باشد. القاء بیماری EAEبا ایمونیزاسیون موش‌های C57BL/6 با تزریق پپتید 55-35 MOG مخلوط شده با ادجوانت کامل فروند صورت گرفت. گروه‌های مبتلا به EAE شامل دریافت کننده‌های ویتامین D3، عصاره زنجبیل، روغن زیتون، بافر فسفات سالین و همچنین PBS به عنوان کنترل تقسیم گردیدند. در روز 31 موش‌های مورد مطالعه کشته شدند و علایم بالینی، شدت التهاب بیماری و تغییرات وزن نیز تا 30 روز مورد بررسی قرار گرفت. داده‌های حاصل توسط آزمون آماری t-test مورد بررسی قرار گرفت.<o:p /> یافته‌ها: گروه کنترل، علائم بالینی را 10روز پس از القاء نشان دادند. این علایم در گروه دریافت کننده ویتامین D3، و عصاره زنجبیل در روز 13 و 15، پس از القاء EAE دیده شد. میانگین حداکثر شدت بیماری در گروه کنترل ویتامین D3(61/1±87/1)، گروه کنترل زنجبیل(79/1±92/1)، گروه ویتامین D3 (01/1±05/1) و در گروه زنجبیل (81/0± 73/0) می‌باشد. ویتامین D3 و عصاره زنجبیل به طور معنی‌داری (05/0=p) باعث افزایش وزن بدن نسبت به گروه کنترل می‌شوند. موش‌های تحت درمان ارتشاح سلول‌های التهابی کمتری را در مقایسه با گروه کنترل نشان می‌دهند.<o:p /> نتیجه‌گیری: نتایج این مطالعه نشان می‌دهد که ویتامین D3 و عصاره زنجبیل می‌تواند در جلوگیری از پیشرفت بیماری و نیز کاهش ارتشاح لکوسیتی در CNS مؤثر باشند.<o:p /> واژه‌های کلیدی: آنسفالومیلیت خود ایمن تجربی، ویتامین D3، عصاره زنجبیل، علایم بالینی<o:p /> Background and Objective: The anti-inflammatory effects of the vitamin D and ginger have been demonstrated in some studies. The aim of this study was to evaluate the effects of vitamin D and ginger extract on the clinical symptoms and leukocyte infiltration of experimental autoimmune encephalomyelitis (EAE).<o:p /> Materials and Methods: For the induction of EAE, the (C57BL/6) were immunized subcutaneously MOG peptide emulsified in complete Freund's adjuvant. The mice divided into EAE group includimg: vitamin D-treated, ginger extract-treated, olive oil and phosphate-buffered saline, and also one group which was considered as normal group and received only PBS(phosphat- Buffered Saline). The mice were sacrificed at day 31 after immunization and the leukocytes infiltration was evaluated in the CNS (central nervous system). The EAE scores and the body weight were evaluated till day 30. Data were analyzed using t-test.<o:p /> Results: Two control groups showed the clinical symptoms at day 10, whereas vitamin D and ginger-treated groups exhibited the symptoms at days 13 and 15, respectively. The maximum mean score of disease was (1.87±1.61), (1.92±1.79), (1.05±1.01), and (0.73±0.81) for olive oil-, PBS-, vitamin D3-, and ginger-treated groups, respectively. Vitamin D3 and ginger extract increased the body weight significantly )P =0.05) as compared to control groups.
