fa بررسی اثر نیکوتین در دوران بارداری و شیردهی بر تغییر بیان لامینین آلفا-5 غشاء پایه در لوله‌های پیچیده دور و جمع‌کننده ادرار در موش Effects of Maternal Nicotine Exposure on Laminin Αlpha 5 Expression Change of the Basal Membrane in the Distal Convoluted and Collecting Tubules in Mice بافت شناسي Histology پژوهشي Research  gte vml 1]><v:roundrect id="_x0000_s1026" style="position: absolute left: 0px text-align: left margin-left: 2.45pt margin-top: 1.4pt width: 449.5pt height: 394.35pt z-index: -1 " arcsize="1475f"> <w:wrap anchorx="page"> </w:wrap></v:roundrect><img alt="AWT IMAGE" width="602" height="528" src="file:///C:/DOCUME~1/ADMINI~1/LOCALS~1/Temp/msohtmlclip1/01/clip_image001.gif" v:shapes="_x0000_s1026" >چکیده<o:p /> زمینه و هدف: در حال حاضر مشخص شده است مادرانی که در دوران بارداری سیگار مصرف می‌کنند، بیان فیبرونکتین و لامینین آلفا-5 در بعضی از ارگان‌های جنین آن‌ها تغییر می‌یابد، به عنوان مثال بیان این پروتئین‌ها در پارانشیم ریه‌های نوزادان کاهش می‌یابد. در مطالعه حاضر، اثر نیکوتین در دوران بارداری و شیردهی بر تغییر بیان لامینین آلفا-5 غشاء پایه در لوله‌های پیچیده دور و جمع‌کننده ادرار در موش مورد بررسی قرار گرفت. <o:p /> مواد و روش‌ها: این مطالعه تجربی بر روی 28 سر موش حامله که به شکل تصادفی به چهار گروه هفت تایی تقسیم شده بودند، صورت گرفت. در گروه آزمایشی روز اول از روز 7 حاملگی تا تولد نوزادان و در گروه آزمایشی روز چهاردهم از روز هفتم بارداری تا 14 روز بعد از تولد نوزادان، نیکوتین به‌ میزان 2 میلی‌گرم بر کیلوگرم و به صورت داخل صفاقی به مادران تزریق شد. در گروه‌های سوم و چهارم، به عنوان گروه‌های کنترل، به ترتیب مشابه با گروه‌های آزمایشی اول و دوم تحت تزریق میزان و مدت زمان معادلی از سالین قرار گرفتند. بعد از آخرین تزریق کلیه همه نوزادان (متوسط 50 نوزاد در هر گروه) بعد از بیهوشی برداشت شده و با استفاده از روش‌‌های ایمنوهیستوشیمی و Real time PCR بیان لامینین مورد بررسی قرار گرفت. داده‌ها با استفاده از آزمون ناپارامتری Mann-Whitney و آزمون t مستقل تجزیه و تحلیل شدند.<o:p /> یافته‌ها: نتایج مطالعه حاضر نشان داد بیان mRNA لامینین آلفا-5 در گروه آزمایش روز اول به میزان 2 برابر در مقایسه با گروه معادل یعنی کنترل روز اول افزایش یافت، اما در گروه آزمایش روز چهاردهم (19/0±62/0) نسبت به گروه کنترل روز چهاردهم (14/0±95/0) کاهش داشت (043/0=p). در روش ایمنوهیستوشیمی در لوله‌های جمع‌کننده گروه‌های آزمایش روز اول و چهاردهم نسبت به گروه‌های کنترل معادل بترتیب افزایش و کاهش معنی‌داری از واکنش مشاهده شد (039/0=p)، در حالی‌که در لوله‌های پیچیده دور تنها اختلاف معنی‌داری از نظر شدت واکنش بین گروه آزمایش روز اول و گروه کنترل روز اول مشاهده شد (041/0=p).<o:p /> نتیجه‌گیری: مصرف نیکوتین توسط مادر با اثر بر روی بیان لامینین به ‌عنوان یکی از ترکیبات مهم غشاء پایه کلیوی، ممکن است یکی از عوامل مؤثر در افزایش بیماری‌های کلیوی در افراد سیگاری باشد. <o:p /> واژه‌های کلیدی: نیکوتین، کلیه، لامینین آلفا- 5، لوله‌های پیچیده دور، لوله‌های جمع‌کننده<o:p />     Background and Objective: It is obviously approved that Fibronectin and Laminin α-5 (LAMA5) expression will change, in some organs of fetuses that their mothers were smoking during pregnancy period. For example, the expression of mentioned proteins in newborns decreases in parenchyma of their lungs. In present study, we evaluated the effects of maternal nicotine exposure on LAMA5 expression change of the basal membrane in the distal convoluted and collecting tubules in Mice.<o:p /> Materials and Methods: The present experimental study was performed on 28 pregnant mice which were randomly divided into four groups (n=7). Pregnant mice in experimental day 1 group received 2 mg/kg nicotine from gestation day 7 (GD7) to last day of pregnancy and experimental day 14 group received the same amount of nicotine from GD7 to post natal day 14(GD14). Control groups 3 and 4 were injected with the same volume of normal saline parallel to experimental groups (Con). After the last injection, all the newborns (average 50 newborns in each group), were anesthetized in order to remove their kidneys and become prepared for Immunohistochemical method and real-time polymerase chain reaction to detect the expression of Laminin. Data were analyzed using non-parametric Man-Whitney U test and t-test.<o:p /> Results: Our results show that LAMA5 mRNA expression in the kidneys of newborns treated with nicotine, Exp D1 increased about 2-fold in comparison with Control day 1 group , but decreased in Exp D14 (0.62±0.19) comparing to Control day 14 (0.95±0.14) (p=0.043). Lama5 Immunoreactivity in collecting tubules respectively increased and decreased in the Exp day1 and day 14 groups in comparison with Control groups (p=0.039).
