fa تأثیر حفاظتی‌ کورکومین‌ و ویتامین‌ D بر تغییرات فیزیولوژیک‌ و هیستوپاتولوژیک‌ بیضه‌ در موش‌های صحرایی‌ بالغ‌ تیمار شده با متوترکسات: یک مطالعه تجربی The Protective Effect of Curcumin and Vitamin D on Physiological and Histopathological Changes of Testis in Adult Rats Treated with Methotrexate: An Experimental Study فيزيولوژي Physiology پژوهشي Research زمینه و هدف: کورکومین و ویتامین D دارای فعالیت آنتیاکسیدانی میباشند. هدف از این پژوهش تعیین اثر حفاظتی کورکومین و ویتامین D بر تغییرات فیزیولوژیک‌ و هیستوپاتولوژیک‌ بیضه‌ در موشهای صحرایی‌ بالغ‌ تیمار شده با متوترکسات بود. مواد و روش ها: در این مطالعه تجربی، 25 سر موش صحرایی نر بالغ به 5 گروه 5تایی تقسیم شدند. گروه کنترل، گروه شاهد: 1 میلیلیتر آب مقطر به عنوان حلال، متوترکسات (٥ میلیگرم/کیلوگرم)، متوترکسات+کورکومین‌ (٢٠٠ میلی‌گرم/کیلوگرم) + ویتامینD (500 واحد بین‌المللی/کیلوگرم)، متوترکسات+کورکومین‌ (4٠٠ میلیگرم/کیلوگرم) + ویتامینD (10٠٠ واحد بین‌المللی/کیلوگرم). پس از 28 روز تیمار با کورکومین و ویتامین D (گاواژ)، خونگیری مستقیم از قلب جهت اندازه‌گیری غلظت سرمی هورمون لوتئینی (Luteinizing hormone; LH)، هورمون محرک فولیکول (Follicle-stimulating hormone; FSH) و هورمون تستوسترون انجام شد. بیضهها خارج و تغییرات بافتی بعد از رنگآمیزی هماتوکسیلین-ائوزین مورد بررسی قرار گرفت. دادهها با استفاده از آنالیز واریانس یکطرفه و آزمون تعقیبی Tukey تجزیه و تحلیل شد. یافتهها: سطح هورمون تستوسترون، تعداد اسپرماتوگونی، اسپرماتوسیت، اسپرم و لیدیگ، در گروه تجربی 1 نسبت به گروههای کنترل و شاهد کاهش معنیداری نشان داد، اما سطح سرمی هورمونهای LH و FSH افزایش معنیداری را نشان داد (05/0>p). سطح هورمون تستوسترون، تعداد اسپرماتوگونی، اسپرماتوسیت، اسپرم و لیدیگ در گروههای تجربی 3 و 4 نسبت به گروه تجربی 1، افزایش معنیداری نشان داد، اما سطح سرمی هورمونهای LH و FSH کاهش معنیداری نشان داد (05/0>p). نتیجهگیری: احتمالاً کورکومین و ویتامین D به دلیل داشتن خاصیت آنتیاکسیدانی و ضدالتهابی قادر است تغییرات فیزیولوژیک‌ و هیستوپاتولوژیک‌ بیضه‌ در موشهای صحرایی‌ بالغ‌ تیمار شده با متوترکسات را بهبود بخشد. واژههای کلیدی: کورکومین‌، ویتامین D، متوترکسات، تستوسترون، بیضه، موش صحرایی   <pre style="text-align:justify"> Background and Objectives: Curcumin and vitamin D have antioxidant activities. The aim of this study was to determine the protective effect of curcumin and vitamin D on the physiological and histopathological changes of the testis in adult rats treated with methotrexate. Materials and Methods: In this experimental study, 25 adult male Wistar rats were divided into 5 groups of 5. Control group, sham group: 1 ml of distilled water as a solvent, methotrexate (5 mg/kg), methotrexate (5 mg/kg) + curcumin (200 mg/kg) + vitamin D (500 IU/kg), and methotrexate (5 mg/kg) + curcumin (400 mg/kg) + vitamin D (1000 IU/kg). After 28 days of treatment with curcumin and vitamin D (gavage), blood was taken directly from the heart to measure serum concentrations of luteinizing hormone (LH), follicle stimulating hormone (FSH), and testosterone. The testicles were removed and tissue changes were examined after hematoxylin-eosin staining. Data were analyzed using one-way analysis of variance and Tukey's post hoc test. Results: Testosterone hormone level, number of spermatogonia, spermatocyte, sperm and Leydig showed a significant decrease in the experimental group 1 compared to the control and sham groups, but the serum level of LH and FSH hormones showed a significant increase (p<0.05). The level of testosterone hormone, the number of spermatogonia, spermatocytes, sperm and Leydig in the experimental groups 4 and 3 showed a significant increase compared to the experimental group 1, but the serum level of LH and FSH hormones showed a significant decrease (p<0.05). Conclusion: Probably, curcumin and vitamin D, due to their antioxidant and anti-inflammatory properties, can improve the physiological and histopathological changes of the testis in the adult rats treated with methotrexate.
